Yes , because, particularly the fruit will affect the reading. Imagine fermenting straight peaches with no water, versus a smaller quantity of peaches in a gallon of water. One stick of vanilla in a gallon isn't going to affect your reading.

Maceration of the fruit is going to give you a better reading on the hydrometer. Makes sure the sugars are fully dissolved into the must. Or at least mostly.
